Recent Advances in DNA Recombination Technology; Construction of Gene Targeting Vector Through Recombineering Technology

Abstract
One of the key technologies in gene targeting is to construct the correct target vector. Recently, target vector construction has been possible without introduction of restriction enzymes and DNA ligases. The underlying principle is to use virus-derived proteins which are involved in the increase of homologous recombination in bacteria. Recombineering is based on the function of bacteriophage proteins exo, bet, and gam, which increase the homologous recombination between template DNA and exogenous DNA in bacteria. This technology will be useful not only in target vector construction, but also in various genome manipulation experiments.
